GastroPlus, developed by Simulations Plus, Inc., is a leading physiologically based pharmacokinetic (PBPK) and mechanistic absorption modeling platform used to predict oral drug absorption, tissue distribution, pharmacokinetics, and pharmacodynamics from discovery through post-approval. It combines the Advanced Compartmental Absorption and Transit (ACAT) model with whole-body PBPK to translate in vitro measurements (such as solubility, permeability, dissolution, and metabolism) and preclinical data into robust human predictions.

Scientists use GastroPlus to guide formulation design, estimate first-in-human doses, assess food and pH effects, evaluate bioequivalence and biowaiver strategies, and simulate special populations (including pediatrics and patients with organ impairment).
